I followed your instruction to disable “Auto detect default locations for saved/…”
But I still cannot find the downloaded file at the specified location when I switch to real host. I try to re-download at the sandbox again. I can see the downloaded file while I chose “save as”. That’s make me confuse.
Addition: I try it at IE8. But the result is same to IE9

Sandbox
The avast! Sandbox is a special security feature that allows you to browse the web, or manually run another application, in a completely safe environment, isolated from the rest of your system.
This is especially useful when visiting potentially infected websites, or if you suspect an application may be infected - you can run the program (or your web browser) inside the sandbox to determine whether or not it is safe, while remaining completely protected against any malicious actions that it may try to carry out.
When you run a browser in the Sandbox all downloads are kept in the Sandbox and removed when the Sandbox is closed.
My question is to use Sandbox for visiting potentially infected websites (may be or not ). And then download the files at the websites. after that, I want to open the downloaded file outside the Sandbox.
By checking the box "automatically detect default locations for saved/downloaded files and save files to these locations outside the sandbox" you can make sure that files saved or downloaded while working in the sandbox will be saved outside the sandbox. In other words, the process of saving or downloading files to these locations will not be virtualized. This box is checked by default, but if unchecked, any files that you save or download while working in virtualization mode will not actually be saved.
This should also work in 6.0.1000. Please try this :
Close IE first, than in the Avast GUI choose : Additional Protection - Sandbox - Expert Settings - Web browsers - be sure ‘Automatically detect default locations …’ is checked - and under Maintenance click ‘delete contents’ - click ‘ok’.
Than try IE sandboxed downloading something again.

I tried already. Even Checked or Unchecked it and then “Delete Contents”, however, it doesn’t work.
Do you have any idea about the location of virtual file will be keep, if the user browsing and download the file from website?
Because when i tried with avast antivirus IS and try to check or uncheck those "Store files in special storage … " and "Automatically detect default locations for saved/download … ", i am still doesn’t able to found the downloaded file on the places that i would like to save that file.
Pardon me to asking, because about this matter till today i am keep looking how to find out the location of virtual folder.
“Automatically detect …” option should do this automatically – when it’s checked on, everything you’re going to download is saved outside sandbox (= is not stored in sandbox, but saved on your disk). It detects default’s web browser download folder and then set an exclusion for the folder. How did you set your download location?
Alternatively, you can set exclusion folder: Expert Settings → Exclusions
