I use all free spyware dectectors. some of these start as trials and will scan all the time. When the trial period ends you have to either buy them or automatic scanning ends. You can then scan your hard drive using them at your discretion. They all have free updates.
Only one scans spyware coming in for free that I know of, and that is

SpywareBlaster doesn’t actually scan for spyware, it blocks known spyware: I reckon it’s a “must” on any system.
If you want to scan for spyware on a system, Ewido, Ad-Aware and Spybot Search & Destroy, as roro said.
If you want to scan downloads to look for Trojans, Ewido as roro said.
If you scan any downloaded files with Ewido, are careful where you download files from, and keep your system up to date (older versions of Windows, IE, Firefox, Quicktime, Sun Java etc allow drive by spyware installation of spyware- where even visiting the wrong site leave you with spyware) you will avoid up to 100% of spyware.
If you take these precautions but still find spyware, you’ll need real time protection.
Spyware Doctor, Sunbelt CounterSpy and Webroot SpySweeper usually come out well in reviews.
A free option with real time protection is Spyware Terminator. It hasn’t appeared in the reviews yet but several people on the forum recommend it.
Microsoft AntiSpyware (beta) has real time protection and is free but has performed poorly in recent reviews.
Personally, I find sensible precautions are enough to avoid spyware, but if you have kids on the computer downloading anything and everything, a good anti-spyware program may be a sound investment.

OK, here’s the short answer:
Install all the programs recommended by roro: they are all free. They all remove spyware.
Update Windows and applications regularly.
Don’t download files from untrustworthy sources, and don’t open email attachments.
If you still get spyware, consider buying Spyware Doctor, CounterSpy or Spysweeper which have real time protection: they block spyware from getting on your system.
If you don’t want to pay, use MS Anti-Spyware or Spyware Terminator: they have real-time protection but you may not get as good protection.
There’s a bit of controversy about this one, but I’ll second I Mazza I’s recommendation.

On demand scanners (scanners which scan for spyware when you choose to run a scan) won’t- they are not in memory until you use them.
You can have Ad-Aware, Spybot and Ewido free on your computer without any slowdown.
Real time scanners (scanners which scan every file as it arrives on your computer or before execution) will, to some extent, because they are always in memory.
If you have a real time scanner like Spyware Doctor, CounterSpy, SpySweeper, the pay version of Ewido, MS AntiSpyware or Spyware Terminator installed, you will notice that they all have running processes in Task Manager.
Whether you notice any slowdown will depend on your system. I’ve tried most on an older computer (256M memory) and not noticed a problem.
All these programs have a free trial, so you can test them to see if they have a negative effect on system performance.
You can have as many on demand scanners installed as you want, but only have one real time scanner running.
If you have more than one real time scanner running at the same time, you may notice slowdowns and conflicts.
