Is it possible to white list a second phone number (Sim card) and, if so, what is the easiest way to do so? (On a Motorola Defy, Android 2.2.2., not rooted).

Yes, you could add other trusted SIM cards.
You can remove (clean) the trusted SIM cards if you need also.
Yes, just insert it into device, put your PIN and the SIM card will be added to trust list.
